Enzyme activities
Enzyme-linked immunosorbent assay (ELISA) was used to measure the enzyme activities of PPAR-α, PPAR-β, LPL, FABP-3, MTP, LXR, ELOVL 5, 6GPD, G6PD and ME with a commercial kit from Shanghai Ding Biological Technology Company Limited (Shanghai, China). A ground glass homogenizer was used to homogenize liver (0.5-1.0 g) on ice. The homogenates were centrifuged (110 000 g, 20 min at 4°C), and the clear phase between the top layer and the pellets was used for the analysis. At 450 nm, an ELISA microplate reader was used to measure the optical density (OD) of samples in a 96-well plate. According to the manufacturer’s instructions, a standard curve was generated and the standard diversity calculated using Excel 2003. Enzyme activity units (IU), defined as moles of substrate, were converted into product per minute at assay temperature.
